Very few such protocols exist for hop. In order to establish a method for the isolation of protoplasts for hop, we tested different media for growth in tissue culture. On the hormone-free medium, growth was fastest and leaves were well developed and suitable for protoplast isolation. By optimizing the composition of the enzyme mixture, we achieved high yield (8.0 × 10^5 protoplasts per g of leaves) and (86 %) viability of the isolated protoplasts in the Celeia cultivar. The established protocol also proved to be suitable for the isolation of protoplasts from other Slovenian hop cultivars, although we observed differences in yield and viability of isolated protoplasts between the cultivars used. We report the first protocol for isolation of protoplasts from hop leaves, which will enable many further applications for hop breeding</dc:description><dc:description xml:lang="sl">Učinkovit protokol izolacije protoplastov je glavni pogoj za razvoj metod transformacije protoplastov, ki imajo široko uporabnost, od bazičnih študij funkcij genov do regeneracije transformiranih rastlin s spremenjenimi lastnostmi. Pri hmelju je takšnih protokolov zelo malo. Z namenom vzpostavitve metode izolacije protoplastov pri hmelju smo preizkusili različna gojišča za rast v tkivni kulturi. Na gojišču brez hormonov je bila ta najhitrejša, listi so bili lepo razviti in primerni za izolacijo protoplastov. Z optimizacijo sestave encimske mešanice smo pri sorti Celeia dosegli visok donos (8,0 × 10^5 protoplastov na g tkiva) in (86 %) živost izoliranih protoplastov. Vzpostavljen protokol se je izkazal za primernega tudi za izolacijo protoplastov pri drugih slovenskih sortah hmelja, pri čemer smo med uporabljenimi sortami opazili razlike v donosu in v živosti izoliranih protoplastov.
